The present application provides a detection chip and a preparation method therefor. The detection chip comprises a substrate material layer; a feature array comprising a plurality of feature units distributed on the substrate material layer; and a feature array protection structure formed on the substrate material layer, the feature array being distributed in the feature array protection structure, wherein the feature array protection structure comprises at least an adsorption material for adsorbing a biological sample. According to the present application, the excitation power enhanced by the detection chip is improved by at least a plurality of orders of magnitude compared with the incident laser power, and with the design optimization and process improvement of the feature array, the enhancement proportion can be further improved, such that the excitation power received by the fluorescent dye is improved without changing the power of the laser, and stronger fluorescence signal intensity is generated.

Provided is a synthesis apparatus for synthesizing biomacromolecules. The synthesis apparatus includes a feeding apparatus, a recognition apparatus, a reaction apparatus, a sorting device, and a transfer device. The feeding apparatus includes a transporting assembly that can simultaneously transporting biochips. The recognition apparatus recognizes an identification of each biochip on the transporting assembly and feed back an identification information to a control device. The reaction apparatus includes various reaction vessels for performing synthesis reactions on the biochips. The sorting device includes various first sorting assemblies located on a side of the transporting assembly and correspond to the reaction vessels. The first sorting assemblies sort the chips on the transporting assembly to the reaction vessels corresponding to the current synthesis reactions when controlled by the control device. The transfer device transfers the biochips in the reaction vessels to the feeding device. Further provided are a synthesizer and a synthesis method.

Methods and compositions for preparing a nucleic acid sequencing library are described including (a)transposing an insertion sequence into first fragments of the target nucleic acid, wherein the insertion sequence comprises a hybridization sequence, and wherein the transposing produces nicks in the first fragments; (b) combining in a single mixture (i) the first fragments of the target nucleic acid from (a), (ii) a splint oligonucleotide, and (iii) a population of beads, wherein each bead comprises capture oligonucleotides immobilized thereon, and (c) ligating capture oligonucleotides of individual beads to inserted hybridization sequences of individual first fragments.

Provided are a method and system for storing information using DNA. The method comprises: 1) obtaining a DNA fragment attribute library, wherein the DNA fragment attribute library comprises a plurality of DNA molecules, and comprises a retrieval table of storage object attributes and the DNA molecules, and each DNA molecule comprises double-stranded DNA, which represents the storage object attribute, and a single-stranded sticky end for connection; 2) on the basis of the retrieval table, encoding information to be stored, so that same is directly mapped to a corresponding DNA molecule in the DNA fragment attribute library; 3) acquiring corresponding DNA fragments from the DNA fragment attribute library, and connecting same, so as to obtain a connected DNA molecule; and 4) storing the connected DNA molecule. The method and system can be extended to different data types and different application scenarios, and prevent an oligonucleotide sequence from being repeatedly synthesized during storage, thereby greatly reducing storage cost.

Provided is a multi-omics library construction method, a detection method and a related kit. The multi-omics library construction method includes: placing a sectioned sample on a chip having a PolyA capture function for protein capture and mRNA capture, and performing protein capture using a nucleic acid-conjugated antibody; performing reverse transcription on captured mRNA and complementary strand synthesis on a nucleic acid sequence of the nucleic acid-conjugated antibody to respectively obtain cDNA and a complementary strand of the nucleic acid sequence; and performing library construction on the cDNA and the complementary strand of the nucleic acid sequence, thereby obtaining a transcriptome library and a proteome library of the sectioned sample.

The method and system of a biochip may automatically schedule and control a plurality of biochips, so as to complete the labeling and capturing of biological information in tissues, and perform overall collection of the captured products, which meet the requirements of high utilization rate and high degree of automation for a biochip.

Provided are a method for constructing a library based on an RNA sample and uses thereof. The method includes: step 1 of subjecting the RNA sample to a reverse transcription reaction to obtain DNA-RNA hybrid strands; step 2 of performing reaction of the DNA-RNA hybrid strands with an endoribonuclease, a first DNA polymerase, a second DNA polymerase, and dATPs to obtain a double-stranded DNA added with dA-tail, where the first DNA polymerase has a 5′-3′ exonuclease activity and a 3′-5′ exonuclease activity, and the second DNA polymerase has no 3′-5′ exonuclease activity; step 3 of ligating the double-stranded DNA added with dA-tail and a sequencing adaptor to obtain a ligated product; and step 4 of subjecting the ligated product to PCR amplification to obtain a sequencing library.

The chip matrix has characteristics such as resistances against high temperature, high humidity and other harsh environments. Meanwhile, by changing pH, surfactant and other components of a solution containing sequences to be sequenced, a surface functional region of the chip matrix can specifically adsorb a sequence to be sequenced.
